A study of the COX-2 inhibitors Celebrex and Vioxx in children ages 6 to 10 is published in the on Feb 10 in. The drug is among the top 10 drugs approved for treatment of pediatric heart failure in the United States. The researchers looked at data from a recent placebo-controlled, multicenter, double-blind, placebo-controlled study, which included 3,848 pediatric patients with heart failure who received celecoxib (Celebrex) or placebo for a mean of two years. Celecoxib was chosen because of its greater efficacy, greater tolerability and fewer adverse reactions than Vioxx, whose COX-2 inhibitors are known to cause gastrointestinal issues. In both children and adults, the rates of death, nonfatal heart attack or renal failure were lower with celecoxib compared to Vioxx, which is known to cause the same side effects. However, the rates of heart failure and renal failure were higher with the two COX-2 inhibitors, and the rates of death were similar between the two groups.
The investigators also noted that children had a higher rate of serious side effects, including gastrointestinal bleeding, with the COX-2 inhibitors compared to Vioxx. In addition, the rates of serious adverse events were higher with the COX-2 inhibitors.
